Long Noncoding RNA-Sox2OT Knockdown Alleviates Diabetes Mellitus-Induced Retinal Ganglion Cell (RGC) injury

Abstract: Retinal ganglion cell (RGC) injury is one of the important pathological features of diabetes-induced retinal neurodegeneration. Increasing attention has been paid to find strategies for protecting against RGC injury. Long noncoding RNAs (lncRNAs) have emerged as the key regulators of many cell functions. Here, we show that Sox2OT expression is significantly down-regulated in the retinas of STZ-induced diabetic mice and in the RGCs upon high glucose or oxidative stress. SOX2OT knockdown protects RGCs against hi… Show more

“…ERG alterations have been observed in streptozotocin (STZ) treated diabetic rats as early as two weeks after the onset of diabetes [55], four weeks in STZ-diabetic mice [56], and three months in the spontaneously diabetic Ins2Akita model [57]. Reduced b-wave amplitude has been reported in STZ diabetic mice, four months after the induction of diabetes [58] and in STZ-induced diabetic rats after seven weeks of diabetes [59]. The neuroprotective effect of MDL 72527 treatment in diabetes-induced retinal dysfunction is confirmed by the ERG analysis showing improved retinal function in diabetic mice.…”

“…Tenon's capsule fibroblast The functional interaction between lncRNA-MEG3 and Nrf2 constitutes the mechanism by which TGF-β2 induces Tenon's capsule fibroblast proliferation after glaucoma filtration surgery via the direct binding of MEG3 to Nrf2 [187] lncRNA-Sox2OT RGCs Sox2OT knockdown plays an antioxidative role via regulating Nrf2/HO-1 signaling activity [188] associated mutations that activate Nrf2 [177,178]. Also, when ROS exceeds the critical threshold, Nrf2 binds to the ARE gene of Klf9 and upregulates Klf9 expression.…”
