Long-Term Amelioration Practices Reshape the Soil Microbiome in a Coastal Saline Soil and Alter the Richness and Vertical Distribution Differently Among Bacterial, Archaeal, and Fungal Communities

Abstract: Globally soil salinity is one of the most devastating environmental stresses affecting agricultural systems and causes huge economic losses each year. High soil salinity causes osmotic stress, nutritional imbalance and ion toxicity to plants and severely affects crop productivity in farming systems. Freezing saline water irrigation and plastic mulching techniques were successfully developed in our previous study to desalinize costal saline soil. Understanding how microbial communities respond during saline soi… Show more

“…PCRs were performed in a 25-μl system containing 12.5 μl PCR premix (Ex Taq TM ; Takara, Shiga, Japan), 0.5 μl (10 μm) of the forward and reverse primers (final concentration of each primer 0.2 pmol μl −1 ), 0.5 μl DNA template (20 ng), and 11 μl sterile double-distilled water. Thermocycle conditions comprised an initial denaturation at 94°C for 10 min, 30 cycles of denaturation at 94°C for 30 s, annealing at 55°C/56°C (16S rRNA/ITS) for 45 s, and extension at 72°C for 1 min, followed by a final extension at 72°C for 10 min ( Sun et al, 2020 , 2022 ).…”

“…Bioinformatic analysis was performed according to previous studies with some modifications ( Shi et al, 2019 ; Sun et al, 2022 ). VSEARCH (version 2.21.1) was used for the bioinformatic analysis of high-throughput sequencing data ( Rognes et al, 2016 ).…”

“…Adoption of traditional amelioration practices on saltaffected coastal lands not only reducesthe direct impact of osmotic stress on crops, but also exerts a positive action on soil biological fertility by reverting the reduction in the biodiversity of the soil microbiome (Sun et al, 2022).…”

“…However, seed germination and crop seedling formation, establishment, and growth are greatly impaired due to the high degree of salinization, nutrient de ciency, low-fertilizer utilization , and poor soil texture and structural stability of mud at soils and the seasonal salt regression and frequent salt accumulation (Dong et al 2020;Odeh and Onus 2008). In some cases, serious osmotic stress and ion toxicity even occur (Sun et al 2022). Therefore, saline-alkaline soil is di cult to use directly because of the restrictive factors in agricultural production (Paul and Lade 2014; Ranasinghe and Piyadasa 2020).…”
