Long-term starvation and ageing induce AGE-1/PI 3-kinase-dependent translocation of DAF-16/FOXO to the cytoplasm

Abstract: Background: The provision of stress resistance diverts resources from development and reproduction and must therefore be tightly regulated. In Caenorhabditis elegans, the switch to increased stress resistance to promote survival through periods of starvation is regulated by the DAF-16/FOXO transcription factor. Reduction-of-function mutations in AGE-1, the C. elegans Class IA phosphoinositide 3-kinase (PI3K), increase lifespan and stress resistance in a daf-16 dependent manner. Class IA PI3Ks downregulate FOXO… Show more

“…For homology-based gene prediction, we aligned Bombyx mori, D. melanogaster, Apis mellifera, A. pisum, Rhodnius prolixus, Tribolium castaneum and Pediculus humanus proteins (from the Ensembl database [23]) to the S. furcifera genome using TblastN [24] with an E-value ≤ 1E −5 , and then used GeneWise2.2.0 [25] for spliced alignment and prediction of gene structures. Secondly, for ab initio prediction, GENSCAN [21] and AUGUSTUS [22] were used to predict genes based on repeat-masked genome sequences.…”

“…The importance of DAF‐18/PTEN tumor suppressor in longevity and dauer larva formation, through the regulation of the IIS pathway, has been established in the late 1990s by several research groups (Ogg & Ruvkun, 1998; Gil et al ., 1999; Mihaylova et al ., 1999; Rouault et al ., 1999). DAF‐18/PTEN has been shown to regulate L1 arrest in the germ line, and insulin‐like signaling appears to be transduced by AGE‐1/PI3K during L1 arrest (Weinkove et al ., 2006; Zhang et al ., 2011b). …”

“…C. elegans loss-of-function mutations in daf -2 have an extended lifespan, which is dependent on the activation of the transcription factor DAF-16 (homologous to human FOXO genes) (Apfeld and Kenyon 1998; Gems et al 1998; Tullet et al 2008). Dietary restriction is one of the most consistent methods for producing lifespan extension in model organisms, activates DAF-16 through a decrease in PI3K/AKT signaling (Weinkove et al 2006). Activation of SIR-2.1, the C. elegans homolog of human SIRT1 also results in lifespan extension through partially overlapping mechanisms, including activation of DAF-16 (Dali-Youcef et al 2007).…”
