Low complexity RGG-motif sequence is required for Processing body (P-body) disassembly

Abstract: P-bodies are conserved mRNP complexes that are implicated in determining mRNA fate by affecting translation and mRNA decay. In this report, we identify RGG-motif containing translation repressor protein Sbp1 as a disassembly factor of P-bodies since disassembly of P-bodies is defective in Δsbp1. RGG-motif is necessary and sufficient to rescue the PB disassembly defect in Δsbp1. Binding studies using purified proteins revealed that Sbp1 physically interacts with Edc3 and Sbp1-Edc3 interaction competes with Edc3… Show more

“…Localization of Sbp1, Scd6 and LSM14 to puncta is dependent on their RGG-motif as the mutants lacking this motif are defective in localizing to puncta 6,7,23 . RGG-motifs are unique as these have been implicated in both assembly and disassembly of RNA granules [24][25][26] . It must be noted that the Lsm domain of Scd6 has the strongest impact on localization of Scd6 to HU-induced puncta (Figure 4A).…”

“…RGG-motifs have been implicated in several cellular processes including translation control. Localization of Sbp1, Scd6 and LSM14A to puncta is dependent on their RGG-motif as the mutants lacking this motif are defective in localizing to puncta 6,7,23 RGG-motifs are unique as these have been implicated in both assembly and disassembly of RNA granules [24][25][26] It must be noted that the Lsm domain of Scd6 has the strongest impact on localization of Scd6 to HUinduced puncta (Figure 4A). The interactions of RGG-motif with RNA and/or proteins that enable localization of these proteins to puncta remain to be explored.…”

“…The current reports on YjeF mainly focus on its function as a domain of Edc3 (enhancer of the mRNA decapping 3). For example, Sbp1 (selenium-binding protein 1) has been reported to directly interact with Edc3 through the YjeF-N domain in yeast [ 20 ]. In Candida albicans, the YjeF domain plays a key role in the Edc3 function.…”
