Low-Dose Tacrolimus Promotes the Migration and Invasion and Nitric Oxide Production in the Human-Derived First Trimester Extravillous Trophoblast Cells In Vitro

Abstract: Placentation is one of the most important determinants for a successful pregnancy, and this is dependent on the process of trophoblast migration and invasion. Progesterone receptors (PGR) are critical effectors of progesterone (P4) signaling that is required for trophoblast migration and invasion conducive to a successful gestation. In immune complicated pregnancies, evidence has shown that abnormal placentation occurs because of aberrant expression of PGR. Therapeutic intervention with tacrolimus (FK506) was … Show more

“…As well described, no chemical treatment such as mitomycin-C to arrest HTR8/SVneo proliferation was needed before the migration/invasion assay. 39 , 40 , 41 , 42 So, after overnight incubation, cells were confluent, and scratch wounds (∼200 μm) were made using the accompanying WoundMaker device (Essen Bioscience). For the invasion assay, the culture medium was aspirated, and 50 μl of GFR Matrigel (5 mg/ml) was added.…”
