LRP-1–CD44, a New Cell Surface Complex Regulating Tumor Cell Adhesion

Abstract: The low-density lipoprotein receptor-related protein 1 (LRP-1) is a large endocytic receptor mediating the clearance of various molecules from the extracellular matrix. In the field of cancer, LRP-1-mediated endocytosis was first associated with antitumor properties. However, recent results suggested that LRP-1 may coordinate the adhesion-deadhesion balance in malignant cells to support tumor progression. Here, we observed that LRP-1 silencing or RAP (receptor-associated protein) treatment led to accumulation … Show more

“…Among these overexpressed candidates (Fig. 1c), 8 out of 12 (along with THBS1) even reach the top 10 %, including genes encoding for CD47 and its co-receptor b1-integrin as well as the endocytic receptor LRP-1, another ligand for TSP-1 that we previously reported as implicated in tumor progression modulation [28,29]. Interestingly, these genes represent the most significant connected genes within THBS1 extended network (Fig.…”

“…LRP receptors and sortilin are both known to play a role in cell adhesion, migration and differentiation3132333435, though as in the case of HSPG, the molecular mechanism underlying their complex functions is far from clear.…”

“…In this way, we recently demonstrated [83] that LRP-1 forms complex with the hyaluronan receptor CD44, which may bind proMMP-9 [24]. The identification of these partners could represent a key to the understanding of the LRP-1 roles in ECM remodeling.…”
