Ocular Inserts of Voriconazole-Loaded Proniosomal Gels: Formulation, Evaluation and Microbiological Studies

Abstract: Background: Voriconazole (VRC) is a triazole broad spectrum antifungal drug, used in the management of versatile fungal infections, particularly fungal keratitis. The obligatory use of niosomal delivery of VRC may reduce the frequency of dosing intervals resulting from its short biological half time and consequently improve patient compliance. Methods: VRC loaded proniosomes (VRC-PNs) were set by the coacervation technique and completely characterized. The developed formula was comprehensively assessed concern… Show more

“…Based on the pattern of the ICs, amorphous materials lose their crystallinity when they undergo complexation, indicating that the OTV has been encapsulated in the cavity of the CDs [44] , [45] . This revealed the OTV molecule is completely amorphized after stable hydrophobic interactions with both the CDs [46] , [47] .…”

“…Finally, a dried extract was obtained by the addition of a small quantity of methanol. It was diluted and measured at an absorbance of 261 nm (El-Emam et al, 2020 ). The following formula was used to calculate the EE%: where Ctotal = theoretical concentration and Cfree = drug concentration in the supernatant fluid.…”

“…26 Alternative administration routes, such as inhalation for VRZ, 27,28 ITZ, 29 and 5FC, 30 have given encouraging results. Eye drops, 31 intrastromal injections, 32–34 and ocular inserts 35 have also been shown to yield satisfactory outcomes for fungal keratitis. New innovative galenic formulations, such as oral formulations of AmB for systemic use (iCo-019, encochleated AmB) 36,37 and liposomal VRZ, 38 are being studied and their impact on TDM must be evaluated.…”
