Lupinalbin A as the most potent estrogen receptor α- and aryl hydrocarbon receptor agonist in Eriosema laurentii de Wild. (Leguminosae)

Abstract: BackgroundEriosema laurentii De Wild. (Leguminosae) is a plant used in Cameroon against infertility and gynecological or menopausal complaints. In our previous report, a methanol extract of its aerial parts was shown to exhibit estrogenic and aryl hydrocarbon receptor agonistic activities in vitro and to prevent menopausal symptoms in ovariectomized Wistar rats.MethodsIn order to determine the major estrogen receptor α (ERα) agonists in the extract, an activity-guided fractionation was performed using the ERα … Show more

“…Flavonoids/isoflavonoids not recorded in that review and/or from different Eriosema species are mentioned below. In a dereplication approach, the flavones isovitexin (3), luteolin (6), isoorientin (7) and luteolin-7-O-glucoside (8) were unambiguously identified in a methanol extract of the aerial parts of E. laurentii (Ateba et al, 2014b) and 2″…”

“…Quercetin (13) and quercetin-3-O-methylether (14) were identified in a methanol extract of the aerial parts of E. laurentii (Ateba et al, 2014b), while 3,4′,6,8-tetrahydroxyflavone-7-C-glucoside (15) from that extract was identified for the first time in the genus (Ateba et al, 2016a).…”

“…In yeast systems expressing the human ERα and the human aryl hydrocarbon receptor, 8 showed agonistic effects with EC 50 values of 21.4 nM and 1.34 μM, respectively. 63 Additionally, 9 was reported to activate ERα with an EC 50 value of 6.1 μM. Regarding 8 and 9 , it needs to be noted that these compounds exert more potent inhibitory effects against 17β-HSD1 than 17β-HSD2.…”
