Lymphatic Senescence: Current Updates and Perspectives

Abstract: Lymphatic flow is necessary for maintenance of vital physiological functions in humans and animals. To carry out optimal lymphatic flow, adequate contractile activity of the lymphatic collectors is necessary. Like in all body systems, aging has also an effect on the lymphatic system. However, limited knowledge is available on how aging directly affects the lymphatic system anatomy, physiology and function. We investigated how senescence leads to alterations in morphology and function of the lymphatic vessels. … Show more

“…The aging process induces changes in structure and function of systemic lymphatic networks 60,[62][63][64] . Muscle cell atrophy, elastic element destruction, and aneurysm-like formations have been identified within aged systemic lymphatic vessels 60,[62][63][64] .…”

Muscle cell atrophy, elastic element destruction, and aneurysm-like formations have been identified within aged systemic lymphatic vessels 60,[62][63][64] . Animal studies have documented a severe decrease in pump action of these vessels with age, mainly due to decreased contractile speed 62,63 , and likely secondary to age-related enlargement of the lymphatic vessels 63 .…”

“…Histological evaluation of the structure of mesenteric lymph nodes in the elderly has revealed general fibrosis, thickening of the capsule and trabeculae, and increased amounts of connective tissue around the blood vessels [36]. The parenchyma of the nodes is typical of involuting lymphoid tissue [43][44][45][46][47][48]. Dense bundles of collagen fibers have been observed in the marginal sinuses of aging lymph nodes, and the sinuses are made of coarse-grained, fragmented fibers and fibroblasts.…”

“…As a result, any malfunction in this system might result in fluid accumulation within the tissue [10]. A chronic and progressive disease, known as lymphedema, may develop [10,11]. Characteristic for this disease are severe tissue swellings, usually occurring in the upper or lower extremities due to a stasis in lymph flow.…”
