Lysosome biogenesis requires Rab9 function and receptor recycling from endosomes to the trans-Golgi network.

Abstract: Abstract. Newly synthesized lysosomal enzymes bind to mannose 6-phosphate receptors (MPRs) in the TGN, and are carded to prelysosomes, where they are released. MPRs then return to the TGN for another round of transport. Rab9 is a ms-like GTPase which facilitates MPR recycling to the TGN in vitro. We show here that a dominant negative form of rab9, rab9 S21N, strongly inhibited MPR recycling in living cells. The block was specific in that the rates of biosynthetic protein transport, fluid phase endocytosis and … Show more

“…Once at the membrane, rabs orchestrate the assembly of membrane fusion machinery (Carr et al 1999). Roles for rabs in vesicle budding and motility have also been described (Echard et al 1998;Riederer et al 1994). In addition, rabs define organelle subdomains by forming distinct clusters in different membrane regions (Sheff et al 1999;Sonnichsen et al 2000;Trischler et al 1999).…”

“…Taken together these data suggest a possible role for rab11 in the budding of a subset of TGN vesicles. Although rab proteins have been primarily associated with vesicletargeting functions, there are a number of examples in which the rab proteins have been suggested to influence vesicle budding (Plutner et al, 1991;Riederer et al, 1994;Jedd et al, 1995Jedd et al, , 1997. Perhaps the most compelling evidence derives from in vitro assays showing a direct role for rab5 in coated vesicle formation at the plasma membrane (McLauchlan et al, 1998).…”

“…Esta Rab participa en el tránsito de proteínas desde la red trans-Golgi y tiene un papel en el reciclamiento del MPR (96,97). A diferencia de lo que ocurre con perlas de látex, se ha observado ausencia en la expresión de Rab9 en las fagosomas que contienen M. tuberculosis luego de una y 24 horas de infección (66).…”
