Mac‐1: a macrophage differentiation antigen identified by monoclonal antibody

Abstract: We have previously described the derivation of M1/70, a hybrid myeloma line secreting monoclonal rat anti‐mouse cell surface antibody (Springer, T., Galfre, G., Secher, D. S. and Milstein, C, Eur. J. Immunol. 1978. 8: 539). We have now investigated the cellular distribution of this antigen using a 125I‐labeled anti‐rat IgG indirect binding assay, the fluorescence‐activated cell sorter, autoradiography and precipitation of cell surface molecules. Screening with a tumor cell panel showed strong reactivity with a… Show more

“…The present study investigates the influence of AMT on PMN-like cells. It is well known that PMN express Gr-1 (Fleming et al, 1993;Hestdal et al, 1991) and CD11b (Hickstein et al, 1987;Ho and Springer, 1982;Leenen et al, 1994;Rosen and Gordon, 1987;Springer et al, 1979), but both markers are not unique to PMN and the relation of both markers to PMN-resembling cells is therefore ambiguous. Previous findings strongly suggest that the Gr-1+ neutrophils are the predominant infiltrating cells in the corneas with HSK (Stumpf et al, 2002;).…”

“…CD11b is the ␣-chain of the ␣ M ␤ 2 or CD11b/CD18 integrin (also known as Mac-1 or Complement receptor 3) and is generally considered a typical myelomonocytic marker involved in intercellular adhesion and phagocytosis (18). It is downregulated when macrophages become resident in many tissues, including the lung (19).…”

“…The sections were pre-treated with peroxide (H 2 O 2 ) to quench endogenous peroxidase activity. MoAbs specifying the following cell surface markers were employed as primary reagents: CD11b [7] (Seralab, Crawley Down, UK), CD4 [8] and CD8 [9] (Becton Dickinson, Moutain View, CA), CD5 [10], CD25 [11] and CD45R (B220) (all from Pharmingen, San Diego, CA), and biotinylated mouse MoAbs to non-polymorphic murine MHC class II (clone 13/4) [12]. Rat MoAb to murine IL-2 (clone S4B6) [13] and interferon-gamma (IFN-) (clone XMG1.2) [14], gifts from Dr T. Mossman (Calgary, Canada), were used to detect the corresponding cytokine-containing cells.…”
