Malaria — how this parasitic infection aids and abets EBV-associated Burkitt lymphomagenesis

Abstract: Burkitt lymphoma (BL) is >90% EBV-associated when this pediatric cancer is diagnosed in regions heavily burden by endemic Plasmodium falciparum malaria and thus has been geographically classified as endemic BL. The incidence of endemic BL is 10-fold higher compared to BL diagnosed in non-malarious regions of the world. The other forms of BL have been classified as sporadic BL which contain EBV in ~30% of cases and immunodeficiency BL which occurs in HIV-infected adults with ~40% of tumors containing EBV. Withi… Show more

“…The CD56 neg CD16 pos NK cells associated with a history of previous P. falciparum infections had poor direct cytotoxicity, lower expression of NK activation markers (NKp46, NKp30 and CD160) and IFNγ, yet expressed higher levels of MIP1β and CD16. This functionally altered CD56 neg CD16 pos NK cell subset was also found in children with endemic Burkitt lymphoma, which is a paediatric cancer associated with chronic P. falciparum and EBV co‐infections . This suggests NK cells are capable of adapting in response to chronic infections and down‐regulate canonical NK cytokine production in favour of ADCC‐mediated cytotoxicity.…”

“…However, larger, multi-site sample sets are needed to truly validate the functional role of host and viral miRNA in eBL. The role malaria plays, either directly or indirectly (Moormann and Bailey, 2016), in aberrant miRNA expression was not addressed within our study but warrants investigation. The dynamic mechanisms by which miRNA target genes and influence cancer progression is clearly an active area of investigation.…”

“…It has an annual incidence of 5–15 cases in 100,000 children in areas experiencing perennial Plasmodium falciparum transmission. Both EBV infection and holoendemic P. falciparum are thought to be etiologically linked to the development of this B cell cancer (reviewed in Moormann and Bailey, 2016). eBL is an aggressive lymphoma that can present in a number of different anatomical locations including the jaw, abdomen, orbital area, central nervous system, and breast or a combination of these sites (Mwanda et al, 2004; Ogwang et al, 2008).…”
