Mangiferin increases Nrf2 protein stability by inhibiting its ubiquitination and degradation in human HL60 myeloid leukemia cells

Abstract: The nuclear factor erythroid 2-related factor 2 (Nrf2)-mediated antioxidant signaling pathway is a key target for cancer chemoprevention. Recent studies have that Nrf2 activation may be the result of an increase in Nrf2 protein stability. Mangiferin (MA), a compound monomer extracted from the mango plant, has antioxidant and cytoprotective activities. Our previous study demonstrated that MA increased Nrf2 expression and activated Nrf2 signaling in hematopoietic cells. Thus, in the present study, we aimed to in… Show more

“…Moreover, the anti-scavenging activity of mangiferin seems to be related to its ability to modulate the Nrf2/ARE (antioxidant response element) signalling detoxification pathway or promote the activation of key detoxifying enzymes [67]. Mangiferin has been proved to modulate Nrf2/ARE signalling pathway in healthy cells by increasing the Nrf2 half-life, the nuclear accumulation and the downstream production of NAD(P)H quinina reductase [67,68]. Many published studies also indicated mangiferin as a new promising anticancer bioactive compound [69] able to inhibit carcinogenesis and cancer cell growth by apoptosis induction both in vitro and in vivo systems [70].…”

“…The half life of QPRT protein was assessed by cycloheximide (CHX) chase assay as described previously 42, 43 . In brief, Huh7.5.1 cells were infected with HCVcc for 48 h, 300 μg/mL CHX was then added to inhibit de novo protein synthesis.…”
