“…The ester motifs are easy to incorporate, chemically stable, bio-cleavable, and their biodegradation kinetics can be effectively controlled [ [189] , [190] , [191] ]. Some examples of ester-based biodegradable lipids include L319 [ 179 , 189 ], YSK12-C4 [ 192 ], CL4H6 [ 193 ], and the Moderna proprietary lipid series [ 126 , 194 ], and Acuitas proprietary lipid series [ 195 ] ( Fig. 6 D).…”

“…223 Recently, siRNA-loaded nanoparticle mediated silencing of signal transducer and activator of transcription 3 and hypoxia-inducible factor 1 a prevented TAM-mediated angiogenesis and decreased tumour size. 224 In vitro, repolarization has also been achieved with nanoparticle delivery of plasmid DNA encoding the IL-12 gene 225 and with overexpression of microRNA-155 in triple negative breast cancer TAMs. 226 Another approach delivers CRISPR-Cas9 gene editing machinery into MĐ€: knock out of signal regulatory protein a, which engages CD47 on cancer cells and prevents phagocytosis, significantly increased MĐ€ targeting of osteosarcoma cells in vitro.…”

“…Since the major challenge for siRNA-based therapeutics is their delivery, Shobaki et al prepared LPNs by mimicking the envelope-type viruses that followed a novel packaging strategy. 59 The LPNs were composed of CL4H6 lipid (a pH-sensitive cationic lipid), in addition to cholesterol (chol) and PEGylated-lipids, which led to a high EE of siRNA (> 90%). Furthermore, the intravenously injection of siRNA-loaded LPNs into the institute of cancer research mice, the LNPs modified with 1,2-distearoylrac-glycero-3-methylpolyoxyethylene (DSG-PEG 2000) at a level of 1 mol-% of total lipid resulted in minimal siRNA leakage and higher integrity and stability of the LNP in the blood circulation.…”
