Map-based cloning and characterization of a gene controlling hairiness and seed coat color traits in Brassica rapa

Abstract: A glabrous, yellow-seeded doubled haploid (DH) line and a hairy, black-seeded DH line in Chinese cabbage (B. rapa) were used as parents to develop a DH line population that segregated for both hairiness and seed coat color traits. The data showed that both traits completely co-segregated each other, suggesting that one Mendelian locus controlled both hairiness and seed coat color in this population. A fine genetic map was constructed and a SNP marker that was located inside a Brassica ortholog of TRANSPARENT T… Show more

“…Recently, a pubescence locus was also been mapped on LG A6 and its responsible gene was show to be a homolog of the Arabidopsis TRANSPARENT TESTA GLABRA 1 (TTG1) gene (Zhang et al 2009). In contrast, Li et al (2009) have recently proposed the involvement of a homolog of the GLABRA1 (GL1) gene in pubescence using a different B. rapa population.…”

“…Pubescence in the Brassica species is an important trait for plant defense against insects (Ågren and Schemske 1992). The presence/absence of pubescence is controlled by a single gene in many cases (Teutonico and Osborn 1994, Song et al 1995, Nozaki et al 1997, Lou et al 2007, Zhang et al 2009, whereas this trait is also reported as a quantitative trait (Ågren and Schemske 1992). However, the degree of pubescence may be controlled by polygenes (Song et al 1995).…”

“…In another study, Lou et al (2007) detected 10 QTLs for leaf traits using three different mapping populations. By synteny analysis of QTL regions of B. rapa with the Arabidopsis genome, Li et al (2009Li et al ( , 2013 identified the leaf lobe depth and leaf hairiness genes, BRASSICA RAPA GIBBERELLIN 20-OXIDASE 3 (BrGA20OX3) AND BRASSICA RAPA GLABRA1, and Zhang et al (2009) successfully cloned a BRASSICA RAPA TRANSPARENT TESTA GLABRA1 gene controlling leaf hairiness and seed coat color. Genetical genomics or expression quantitative trait locus (eQTL) mapping entails a QTL analysis of highthroughput transcript expression patterns in a genotyped population.…”

“…The SRAP technique was used for several purposes, including genetic map construction (Li et al, 2003;Gao et al, 2007;Sun et al, 2007), QTL mapping Fu et al, 2007;Zhang et al, 2009b), gene tagging and cloning (Rahman et al, 2007;Yi et al, 2008;Zhang et al, 2009a), and genetic diversity (Riaz et al, 2001;Budak et al, 2004a,b;Vandemark et al, 2006).…”
