Plants bind solar energy to organic matter via photosynthesis and assimilation of carbon dioxide from the atmosphere and comprise the major source of nutrition and bioenergy. Plant biotechnology contributes to solution of important constraints in food and feed production and creates new technologies and applications for the sustainable use of plant resources. Genome-wide approaches such as massive parallel sequencing and microarrays to study gene expression, molecular markers for selection of important traits in breeding, characterization of genetic diversity with the aforementioned approaches, and somatic hybridization and genetic transformation are important tools in plant biotechnology. In this paper, studies carried out on enhanced resistance to viruses and tolerance of cold stress in potato, genetic modification of flower pigmentation and morphology in gerbera, production of edible vaccines in transgenic barley seeds, and expression of heterologous proteins for pharmaceutical purposes from vector viruses were chosen to exemplify the general utility of biotechnological approaches and also how plant biotechnology research has developed on cultivated plants at University of Helsinki. The studies reveal cellular and genetic mechanisms and provide scientific information that can be used for widening the uses of crop plants. They can also be used to detect any putative risks associated with the use of the biotechnological application in agriculture and horticulture and to develop practises which reduce any inadvertent negative consequences that plant production may have to the environment.

The most important viral pathogens of the cultivated potato are Potato virus X (PVX) and Potato virus Y (PVY), which can reduce potato production up to 80%. Thus resistance breeding is one of the major goals of plant breeders. Wild potato species are good sources of resistance (R) genes. The resistant plants respond to viral infection with hypersensitive reaction (HR) or extreme resistance (ER). HR is accompanied by programmed cell death, while ER localizes the virus at the primary infection site and limits virus replication without visible symptoms. While HR is generally strain-specific, ER can act against a broad spectrum of viral pathogens. This review aims to describe the molecular mechanisms of resistance against PVX and PVY in potato.

Resistance gene-like fragment ADG2 has been shown to co -segregate with Ryadg, which controls extreme resistance to potato Y potyvirus (PVY) in Solanum tuberosum subsp. andigena (Hamalainen et al. 1997(Hamalainen et al. , 1998. Applicability tests of ADG2 as a selection tool for resistance to PVY were carried out by genomic Southern analysis on a broad range of cultivars and breeding lines of potato and some other solanaceous species. A new marker band (10.5 kb associating with Ry was found in addition to previously reported 3.5 kb marker band. While the 3.5 kb marker was specifically linked to Ryadg with extremely high association (96 %), the newly identified 10.5 kb marker band was detected in genotypes containing Ryadg and other Ry genes. Some signals with ADG2 were also observed in other species within family Solanaceae inferring with the diversity of the corresponding chromosome region(s) among solanaceous species.
