Marked Improvement in Photoinduced Cell Death by a New Tris-heteroleptic Complex with Dual Action: Singlet Oxygen Sensitization and Ligand Dissociation

Abstract: The new tris-heteroleptic complex [Ru(bpy)(dppn)(CH3CN)2](2+) (3, bpy = 2,2'-bipyridine, dppn = benzo[i]dipyrido[3,2-a;2',3'-c]phenazine) was synthesized and characterized in an effort to generate a molecule capable of both singlet oxygen ((1)O2) production and ligand exchange upon irradiation. Such dual reactivity has the potential to be useful for increasing the efficacy of photochemotherapy drugs by acting via two different mechanisms simultaneously. The photochemical properties and photoinduced cytotoxicit… Show more

“…The dual-active compound cis -[Ru(bpy)(dppn)(CH 3 CN) 2 ] 2+ (Figure 5) absorbs efficiently in the visible region due to Ru→bpy and Ru→dppn prox 1 MLCT transitions. 107 Following 1 MLCT state formation, thermal population of the dissociative 3 LF state and the 3 ππ* can occur competitively, leading to either exchange of CH 3 CN for solvent (Φ 400 = 0.002(3) in H 2 O) or production of 1 O 2 (Φ 460 = 0.72(2) in MeOH), respectively. The complex is cytotoxic toward HeLa cells when irradiated with 466 ± 20 nm light with an IC 50 value of 0.47±0.02 μM and a phototoxicity index, PI, of 1110 (PI = dark IC 50 /irr IC 50 ).…”
