Matrix Metalloproteinase-9 −1562C/T Gene Polymorphism Is Associated with Diabetic Nephropathy

Abstract: To investigate the association between the metalloproteinase-9 (MMP9) −1562C/T polymorphism and diabetic nephropathy (DN) in Han Chinese, the patients with type 2 diabetes were collected and divided into the non-DN (NDN) and DN groups; controls were recruited. Genotype and allele frequencies were assessed using polymerase chain reaction and restriction fragment length polymorphism. Results showed that SBP, DBP, HbA1c, UAER, Cr, BUN, TG, and TC were higher in the DN group compared with the control and NDN group… Show more

“…For amplification of the region containing the MMP-9 −1562C/T polymorphism, the following primers were used: sense primer 5-CTT CCT AGC CAG CCG GCA TC-3 and antisense 5-GCC TGG CCT ATA GTA GGC CC-3 [13]. PCR amplification was performed using DreamTaq PCR Green Master Mix (2X) (cat no k1081, Lithuania, EU).…”

“…PCR amplification was performed using DreamTaq PCR Green Master Mix (2X) (cat no k1081, Lithuania, EU). PCR was performed using the following thermal cycling conditions [13] Digestion of PCR products with SphI restriction endonuclease Thermo Scientific FastDigest, cat no FD 0604, Lithuania, EU, to detect the MMP-9 −1562C/T SNP [13]. Separation of restriction products by electrophoresis in 2% agarose gels, after that visualization of the restriction products using an ultraviolet transilluminator.…”

“…Moreover, MMPs, including MMP9 , are zinc-dependent endopeptidases and the major proteases in ECM degradation. There are common variants such as rs3918242 (-1562C/T) and microsatellites (CA)n in the promoter region and several SNPs rs481480, rs2032487, rs4281481, rs3752462 and rs3918242 are found to be associated with the susceptibility to DKD (Hirakawa et al, 2003; Nair et al, 2008; Ahluwalia et al, 2009; Freedman et al, 2011; Cooke et al, 2012; Zhang et al, 2015; Feng et al, 2016).…”

“…A series of epidemiological researches have assessed the association between the MMP-9 gene polymorphism and the risk of DN. [ 25 27 ] The most common variants are microsatellites (CA)n [ 28 ] and rs3918242 (-1562C/T) [ 26 , 27 ] promoter polymorphisms. A recent meta-analysis provided potential evidence of the association between MMP-9 gene promoter polymorphism and the risk of diabetic microvascular complications, [ 26 ] but there is no system review specifically evaluating MMP-9 SNPs in DN to date.…”
