Measurement of Complement Proteins C2 and B in Systemic Lupus erythematosus and Septic Shock

Abstract: We developed sensitive radioassays to quantitate the homologous complement proteins C2 and B, in intact form, in sera from patients with diseases associated with hypo- complementemia. In a study of systemic lupus erythematosus (SLE) patients, we found a subset of patients (6/40) with low C2 concentrations, though overall, C2 concentration was not related to disease activity. Protein B concentration was increased in 23/40 SLE patients, apparently not due to a generalized acute phase response. Among patients wit… Show more

“…In these patients, the alternative pathway appears to be activated early in septic shock, whereas the classical pathway is activated later. Oglesby et al also reported that among septic shock patients (both Gram-negative and Gram-positive) with preexisting cirrhosis, factor B levels were decreased [34]. Lin et al reported that the active fragment of factor B, Bb as well as the Bb to factor B ratio, was significantly increased in septic shock patients [29].…”

“…To quantify the C2, factor B and factor D secreted by U-105-MG cells, solid-phase RIMAs were performed as previously described [21,22]. All three RIMAs utilize rabbit polyclonal antibodies in the solid phase and 125I-labelled mouse mAb as reporters.…”

“…Serum from ambulatory SLE patients was collected on rou tinely scheduled visits to the University of Alabama Clinics. The clinical parameters of these individuals have been previously described [12], All the SLE patients were female, and none had severely active disease based on the scale of Valentijn et al [32] which includes physical examination and laboratory studies (complete blood count, platelet count, blood urea nitrogen, creatinine, and urinanalysis). No pa tient had more than three organ systems involved.…”

“…The wide range of C4BP serum concentrations in nor mal individuals is not unusual when com pared to other complement proteins. For ex ample, complement protein D [38], Clq, Cls, C3, C4, C5, C9 [39], C2, and B [12], all have wide ranges in normal serum.…”
