Measuring long‐chain acyl‐coenzyme A concentrations and enrichment using liquid chromatography/tandem mass spectrometry with selected reaction monitoring

Abstract: Long chain acyl-CoA's (LCACoA) are the activated form of long chain fatty acids and serve as key lipid metabolites. Excess accumulation of intracellular LCACoA, diacylglycerols (DAGs) and ceramides may create insulin resistance with respect to glucose metabolism. We present a new method to measure LCACoA concentrations and isotopic enrichment of palmitoyl-CoA ([U-13C]16-CoA) and oleoyl-CoA ([U-13C]18:1-CoA) using UPLC/MS/MS to quantitate 7 different LCACoA (C14-CoA, C16-CoA, C16:1-CoA, C18-CoA, C18:1-CoA, C18:… Show more

“…All analytes were monitored as their [M Ϫ H] Ϫ ions. LCFa-CoA esters were estimated using the LC-MS/MS method (9). After extraction in the presence of internal standard (20 ng of heptadecanoyl-CoA), samples were analyzed by UHPLC-ESI-MS/ MS operating in multiple reaction monitoring mode [Waters Acquity UHPLC, C8 UPLC BEH column 2.1 ϫ 150 mm, 1.7 m (Waters, Milford, MA) and TSQ Quantum Ultra triple-quad mass spectrometer (Thermo Fisher Scientific, Waltham, MA)].…”

“…The highest amount of propionyl-CoA was also found in the kidney. Saturated acyl-CoAs with a carbon chain length >C 10 were in the highest abundance in the heart. The brain Acyl-CoAs possess structural diversity to include saturated, unsaturated, hydroxylated, and dicarboxyl functional groups in the acyl chain.…”

“…Interestingly, the kidney has relatively higher amounts of other short-and medium-chain saturated acyl-CoAs (C 3 to C 10 ) compared with other organs tested. Odd carbon number acyl-CoAs, such as heptanoyl-CoA and pentanoyl-CoA, are uncommon acyl-CoAs.…”

“…The analytical column was a reverse-phase Zorbax SB-C8 day-to-day variation, and can detect concentration and enrichment in as little as 20 mg of muscle. Measuring both concentration and isotopic enrichment of DAG and other lipid groups ( 11,12 ) can help to elucidate the contribution of plasma FFA to intramyocellular fatty acid-containing compounds. We believe that this method can be used to elucidate the dynamics of the intramyocellular DAG pool and its relationship to plasma FFA in physiological and pathological states.…”
