Mechanical, In vitro Antimicrobial, and Biological Properties of Plasma-Sprayed Silver-Doped Hydroxyapatite Coating

Abstract: Implant related infection is one of the key concerns in total joint hip arthroplasties. In order to reduce bacterial adhesion, silver (Ag) / silver oxide (Ag2O) doping was used in plasma sprayed hydroxyapatite (HA) coating on titanium substrate. HA powder was doped with 2.0, 4.0 and 6.0 wt% Ag, heat treated at 800 °C and used for plasma spray coating using a 30 kW plasma spray system, equipped with supersonic nozzle. Application of supersonic plasma nozzle significantly reduced phase decomposition and amorphou… Show more

“…23 This is probably, on the one hand, due to the constraints of the fivemembered ring that do not allow the optimum orientation of the lone pair on the nitrogen atoms, and, on the other hand, due to the larger basicity of the secondary amine. All of the remaining amino acids are predicted to coordinate in a tridentate fashion with side chain participation (5). In these cases, the silver-coordination bonds with the termini are elongated as compared to the bidentate structures of the unpolar, aliphatic amino acids (by ca.…”

“…Moreover, it has to be taken into account that excessive silver content within hydroxyapatite may be toxic, not only towards microorganisms but also towards mammalian tissues, and may lead to the inhibition of their growth and development and to their death in the case of too high Ag + amounts [190,207,208], toxic silver concentrations being in the range 1-3 μg/ml [191]. Thus, it is fundamental to select the silver doping amount in order to effectively kill microorganisms, without adversely influencing the mammalian tissue condition.…”

“…47 Accordingly, silver released from the Ag-PIII samples to PBS after a 30-day incubation was evaluated by inductively coupled plasma mass spectrometry. As shown in Figure 4, the amounts of silver released from 30 min-15 Ag-PIII, 60 min-15 Ag-PIII, and 90 min-15 Ag-PIII were 3.6 ppb, 4.3 ppb, and 4.6 ppb, respectively, signifying that the leakages of silver from Ag-PIII samples were minimal.…”
