“…For example, both BMP and TGF-β signaling increase baseline cellular contractility (Bhadriraju et al, 2007). Our data show that TGF-β expression is highly upregulated with dynamic stretch at early time points and while TGF-β in the media did not reach detectable levels, we did observe increased Smad3 signaling in response to stretch (Figure 6—figure supplement 1e, [Furumatsu et al, 2013; Li et al, 2015]). Thus, elements of the TGF-signaling pathway may be mechano-responsive, in a ligand or ligand-less fashion (Heo et al, 2016).…”

“…When the cells reached 80%-90% confluence, they were washed with PBS, detached by 0.25% trypsin, and subcultured in new 25 cm 2 flasks at 1 × 10 4 cells/cm 2 . Cells were collected at the second or third generation and sent for flow cytometry analysis (BD, Franklin Lakes, NJ, USA) of CD29, CD34, CD44, and CD45 expression for confirmation of BMSC phenotype [13,14].…”

“…Previous studies have shown that lack of exercise and the associated reduction in stem cell mechanical stimulation decreases osteogenic differentiation and increases the risk of osteoporosis [11,12]. Indeed, our previous studies have demonstrated that mechanical stress can promote osteogenic differentiation and inhibit adipogenic differentiation of BMSCs [13,14]. Further understanding of how mechanical stimuli, especially mechanical stress, affect skeletal tissue differentiation will provide insight into bone repair processes that may be exploited for novel therapeutic strategies.…”

“…Here, the authors found that by pretreating MSCs with TGFβ1/Smad2 pathway antagonists suppressed this increase in Smad2 phosphorylation, whereas pretreatment with TGFβ1/Smad2 signalling agonists facilitated the inhibitory effect of stretch on the adipogenic differentiation markers, suggesting that the anti-adipogenic effects of stretch are mediated in some way by the activation of the TGFβ1/Smad2 signalling pathway [42]. …”
