Mechanically Tunable Hollow Silica Ultrathin Nanoshells for Ultrasound Contrast Agents

Abstract: Perfluoropentane (PFP) gas filled biodegradable iron-doped silica nanoshells have been demonstrated as long-lived ultrasound contrast agents. Nanoshells are synthesized by a sol-gel process with tetramethyl orthosilicate (TMOS) and iron ethoxide. Substituting a fraction of the TMOS with R-substituted trialkoxysilanes produces ultrathin nanoshells with varying shell thicknesses and morphologies composed of fused nanoflakes. The ultrathin nanoshells had continuous ultrasound Doppler imaging lifetimes exceeding 3… Show more

“…This phenomenon is typically used for ultrasound‐based diagnostic imaging. As a widely employed diagnostic‐imaging modality clinically, US imaging is featured with real‐time observation, noninvasiveness, high efficiency, low cost and high safety . In addition to typical imaging capability, US also has found broad therapeutic applications, such as US‐triggered drug releasing and focused ultrasound‐induced hyperthermia (Figure ) .…”

“…Milli-Q purified water was obtained from a Millipore SuperQ Plus Water Purification System (Billerica, MA). The 2-lm microshells used in this study were synthesized with methods previously developed by Liberman et al 27 To visualize transition of the particles, an MR contrast agent, Gd 2 O 3 , was incorporated into the PFP liquid. Gadolinium oxide nanoparticles (2-nm diameter) were manufactured following a modified polyol method.…”

“…Silica‐shell ultrasound‐triggered phase‐shift emulsions (sUPEs) can be transitioned to microbubbles to enhance the effects of therapeutic ultrasound and act as multimodality contrast agents . sUPEs also exhibit increased stability and longevity in vivo compared with microbubbles or other nanoparticle formulations . Transition of sUPEs containing MR contrast agents may increase MR image intensity for clearer delineation of the ablation zone .…”
