Mechanisms involved in A2E oxidation

Abstract: A2E is one of the bis-retinoid pyridinium compounds that accumulate as lipofuscin pigments in retinal pigment epithelial (RPE) cells in association with aging and in some inherited forms of retinal degeneration. Here we observed that 430 nm irradiation of A2E in the presence of the spin trap DMPO, led to the appearance of a superoxide dismutase-inhibitable electron paramagnetic resonance (EPR) spectrum characteristic of DMPO-OH; this finding was indicative of hydroxyl radical (OH) formation following initial s… Show more

“…2, A-D). Moreover, although production of oxidized A2E from A2E had been reported for in vitro systems (6,10,11,39,42), amounts of A2E did not change after illumination (Fig. 2D), suggesting that neither A2E nor oxidized A2E causes light-induced acute retinal degeneration in Rdh8 Ϫ/Ϫ Abca4 Ϫ/Ϫ mice.…”

“…Further evidence comes from the inhibitory effects of known quenchers/ scavengers of singlet oxygen (14). At least in the case of A2E, electron spin resonance spectroscopy indicates that superoxide anion is also generated following 430 nm irradiation of the compound (15). The oxygen-containing moieties ensuing from the photoexcitation of A2E and all-trans-retinal dimer include epoxides, furanoid oxides, and cyclic peroxides (11-13, 16, 17).…”

“…3 possess fluorescent properties and are the source of fundus autofluorescence (Sparrow et al, 2010b;2010c). Moreover, it seems that these bis-retinoids form by the random condensation reactions of two molecules of all-transretinal, and are highly susceptible to photooxidation Sparrow et al, 2002;Dillon et al, 2004;Fishkin et al, 2005;Jang et al, 2005;Wang et al, 2006;Kim et al, 2008;Wu et al, 2010). RPE cells, different from other types of cells, are constantly exposed to sunlight strongest at 400-700 nm.…”
