Medium-term and long-term in vitro conservation and safe international exchange of yam (Dioscorea spp.) germplasm

Abstract: Yam edible tubers feed million of peoples in the intertropical area, where they represent 12% of human feeding. However, as a vegetatively propagated crop, yam is seriously affected by an accumulation of pathogens. Establishing in vitro germplasm collection is a process that cleans the plants from all diseases but viruses. It gives a good control on the preservation of the yam genetic resources and facilitates international exchanges of healthy plant material. Two kinds of in vitro germplasm preservation were c… Show more

“…Pertumbuhan tinggi tanaman pada media MS yang mengandung manitol 40 dan 50 g/l sampai umur 24 minggu masih lambat sehingga belum diperlukan subkultur. Interval subkultur yang lama akan menghemat biaya, mengurangi laju kontaminasi dan menghindarkan terjadinya mutasi pada plasma nutfah yang disimpan (Malaurie et al, 1998).…”

“…Sucrose concentrations range from 0.3 to 1 M and treatment is for 16-4 weeks. In the encapsulation-dehydration protocol sugar concentrations up to 1 M are applied (Malaurie et al 1998a). For lily (Bouman et al 2003) and garlic (Kim et al 2004a), cold acclimation and sugar hardening are applied simultaneously.…”

“…The encapsulation-dehydration method was successfully applied to Cynodon, Zoysia, and Lolium grasses ), sugarcane (González-Arnao et al 1993Paulet et al 1993), yam (Malaurie et al 1998a;Malaurie et al 1998b;Mandal et al 1996), lily and banana (Panis 1995) After encapsulation in alginate beads, meristems are exposed to liquid medium with 0.75 M sucrose. In yam, sugar concentrations are increased step-by-step to 0.9 or 1 M, then beads are dehydrated in a container with dry silica gel or in the laminar airflow bench to achieve moisture contents between 19% and 26% and then plunged into LN (Chap.…”
