Melatonin alters neuronal architecture and increases cysteine‐rich protein 1 signaling in the male mouse hippocampus

Abstract: Melatonin is a lipophilic endogenous indolamine primarily produced by the pineal gland and secreted at night (Ng, Leong, Liang, & Paxinos, 2017). Its main function is to synchronize the body's biological rhythms (Valdes-Tovar et al., 2018), and its pleiotropic effects include a variety of neuroendocrinal and neurophysiological processes (Liu et al., 2016; Ma, Sun, et al., 2017). The peripheral and core activities of melatonin are defined by its intrinsic antioxidant proper

“…Golgi staining was used to visualize the dendritic branching complexity and spines of the neurons in the mice hippocampi. The methods and protocols employed were previously described [ 30 , 58 ]. The FD Rapid Golgistain™ Kit (FD Neurotechnologies, Ellicott City, MD, USA) was used, according to the manufacturer’s instructions.…”

“…The methods for quantifying dendritic complexity, dendritic length, and the number of branch points were described for our previous studies [ 30 , 58 ] with some modifications. Briefly, Sholl’s concentric method [ 59 ] used with the ImageJ (National Institutes of Health, Bethesda, MD, USA) program was used to analyze the neuronal tracings visualized by the 200× Leica DM750 optical microscope (Leica Microsystems, Wetzlar, Germany) and traced using the Leica Application Suite (version 4.12, Leica Microsystems) and Adobe Photoshop CS6 (Adobe Systems, San Jose, CA, USA).…”

“…Immunohistochemical analysis of free-floating sections was performed as per protocols described in previous studies [32][33][34]. After sacrificing the animals, brain tissues were collected and fixed with 4% (w/v) paraformaldehyde in phosphate-buffered saline (PBS).…”
