Mesenchymal stem cell transplantation in tight-skin mice identifies miR-151-5p as a therapeutic target for systemic sclerosis

Abstract: Systemic sclerosis (SSc), an autoimmune disease, may cause significant osteopenia due to activation of the IL4Rα/mTOR pathway. Mesenchymal stem cell transplantation (MSCT) can ameliorate immune disorders in SSc via inducing immune tolerance. However, it is unknown whether MSCT rescues osteopenia phenotype in SSc. Here we show that MSCT can effectively ameliorate osteopenia in SSc mice by rescuing impaired lineage differentiation of the recipient bone marrow MSCs. Mechanistically, we show that donor MSCs transf… Show more

“…66 In addition, miR-151, a direct inhibitor of the mammalian target of rapamycin (mTOR) pathway, when transferred by MSC EVs, has been shown to ameliorate systemic sclerosis. 67 The natural origin of EVs, along with their high stability in the circulation, makes them ideal candidates for targeted therapy by efficient transfer of molecules upon engineering. 68 The strategy of engineering EVs with pro-regenerative molecules is currently gaining an increasing interest.…”

“…Primary bone marrow SSCs (BMSSCs) from female mice were prepared by flushing the femur/tibiae marrow cavity as previously described ( 14 ). Briefly, 15 × 10 6 cells were seeded in 100-mm dishes (Genesee Scientific).…”

“…Interestingly, expansion of SSCs occurs early in the healing microenvironment ( 13 ), suggesting a possible interaction between SSCs and inflammatory cells. SSCs have demonstrated a potent immune-modulatory function in vitro and have been used to treat symptoms of inflammatory diseases ( 14 , 15 ). However, isolation and in vitro expansion of SSCs for transplantation use is artificial and does not accurately represent a potential in vivo function of SSCs.…”
