Mesenchymal Stem Cells Extract (MSCsE)-Based Therapy Alleviates Xerostomia and Keratoconjunctivitis Sicca in Sjogren’s Syndrome-Like Disease

Abstract: Sjogren’s syndrome (SS) is an autoimmune disease that manifests primarily in salivary and lacrimal glands leading to dry mouth and eyes. Unfortunately, there is no cure for SS due to its complex etiopathogenesis. Mesenchymal stem cells (MSCs) were successfully tested for SS, but some risks and limitations remained for their clinical use. This study combined cell- and biologic-based therapies by utilizing the MSCs extract (MSCsE) to treat SS-like disease in NOD mice. We found that MSCsE and MSCs therapies were … Show more

“…AQP5 has been used as a salivary acinar cell marker and displayed elevated gene expression in SGs of NOD mice treated with either spleen or mesenchymal stem cells [ 175 , 176 ]. Elevated gene expression of AQP1, AQP4 and AQP5 have been measured in mice models of xerostomia treated with a cell extract from either bone marrow or mesenchymal stem cells, which reduced salivary focus score, and restored saliva and tear flow rates [ 177 , 178 , 179 ].…”

“…These findings showed that transplanted MSCs may have exerted their regenerative effect either via differentiation of MSCs to SGs epithelial cells or promotion of stem cell of submandibular gland. The expression of Ki-67 as one of the several salivary and lacrimal glands markers involved in regeneration and proliferation has been recently demonstrated by Abughanam et al. (2019) .…”

“…Mesenchymal stem cells (MSCs) are multipotent stromal cells that possess self-renewing capacity and give rise to many specialized cell types such as chondrocytes, adipocytes, myocytes, and osteoblasts. MSCs are primarily located within the bone marrow (BM) stroma, but they can be isolated from almost any organ [ 86 , 87 , 88 ]. MVs produced by horse amniotic MSCs reduced the secretion of TNF-α and, to a certain extent, TGF-β and IL-6 from primary equine alveolar macrophages [ 89 ].…”
