Mesenchymal stem/stromal cells precondition lung monocytes/macrophages to produce tolerance against allo- and autoimmunity in the eye

Abstract: Intravenously administered mesenchymal stem/stromal cells (MSCs) engraft only transiently in recipients, but confer long-term therapeutic benefits in patients with immune disorders. This suggests that MSCs induce immune tolerance by long-lasting effects on the recipient immune regulatory system. Here, we demonstrate that i.v. infusion of MSCs preconditioned lung monocytes/macrophages toward an immune regulatory phenotype in a TNF-α-stimulated gene/protein (TSG)-6-dependent manner. As a result, mice were protec… Show more

“…Consistent with our previous observations with MSCs (Ko et al., 2016, Kota et al., 2013), MSC-derived EVs suppressed Th1 development and inhibited activation of APCs and T cells. In addition, they also increased expression of the immunosuppressive cytokine IL-10 and suppressed Th17 cell development.…”

“…The decrease in BAL cell CD8 and CD21 expression by hAECs demonstrates modulation of airway inflammation in ventilated lambs. Our observations are in agreement with a recent study showing mesenchymal stem cell (MSC)-mediated immune modulation in the airways involving monocytes/macrophages [42]. …”

“…The mechanism of action of MSCs in suppressing T cell functionality goes beyond that of IDO and includes other synergistic responses, including PD-L1/L2 and likely others (Chinnadurai et al, 2014). IFNγ stimulation of MSCs faithfully recapitulates the molecular genetic changes that are observed in MSCs co-cultured with activated PBMCs, except for a few, such as TNF-stimulated gene 6 protein (TSG-6), IL-8 (which could be activated through TNF-α), IL-1, or other PBMC-derived cytokines independent of IFNγ (Fan et al, 2012; Ko et al, 2016; Prockop, 2013; Yun et al, 2017). In conclusion, interrogating the secretome and transcriptome dynamic response of MSCs to defined morphogen or leukine cues may well serve as the best means of interrogating the fitness and potency of MSC products for advanced clinical trials.…”
