Metabolic Reprogramming in Response to Alterations of Mitochondrial DNA and Mitochondrial Dysfunction in Gastric Adenocarcinoma

Abstract: We used gastric cancer cell line AGS and clinical samples to investigate the roles of mitochondrial DNA (mtDNA) alterations and mitochondrial respiratory dysfunction in gastric adenocarcinoma (GAC). A total of 131 clinical samples, including 17 normal gastric mucosa (N-GM) from overweight patients who had received sleeve gastrectomy and 57 paired non-cancerous gastric mucosae (NC-GM) and GAC from GAC patients who had undergone partial/subtotal/total gastrectomy, were recruited to examine the copy number and D3… Show more

“…Unlike normal cells, which use oxidative phosphorylation in mitochondria for energy, GC cells also predominantly use the glycolytic pathway 53 . However, other studies have also proposed that mitochondrial respiration and glycolysis pathways significantly influence the proliferation, 54,55 invasion, 56,57 and metabolic reprogramming 58 behaviors of GC cells. DLAT, as part of the PDC complex, regulates the production of acetyl‐CoA linking glycolysis with the TCA cycle.…”

“…In addition to mutations that directly affect mtDNA, mutations in nDNA-encoded mitochondrial enzymes have been identified in certain carcinomas [4,58]. Despite the extensive research on the role of mitochondria in cancer formation and progression, there is still much to be learned about this process.…”

“…However, the light strand only codes for 8 tRNAs and one subunit. Despite that mitochondrial DNA does not code for a great quantity of RNAs or proteins, it is essential for good cell functionality [19][20][21].…”
