Metabolite Generation via Microbial Biotransformations with Actinomycetes: Rapid Screening for Active Strains and Biosynthesis of Important Human Metabolites of Two Development-Stage Compounds, 5-[(5S,9R)-9-(4-Cyanophenyl)-3-(3,5-dichlorophenyl)-1-methyl-2,4-dioxo-1,3,7-triazaspiro[4.4]non7-yl-methyl]-3-thiophenecarboxylic Acid (BMS-587101) and Dasatinib

Abstract: ABSTRACT:The enzymes present in many microbial strains are capable of carrying out a variety of biotransformations when presented with drug-like molecules. Although the enzymes responsible for the biotransformations are not well characterized, microbial strains can often be found that produce metabolites identical to those found in mammalian systems. However, traditional screening for microbial strains that produce metabolites of interest is done with many labor intensive steps that include multiple shake flas… Show more

“…The enzymes that are present in many microbial strains allow access to a wide range of biotransformation products. This approach, which is especially used in the preparation of mammalian metabolites (Smith and Rosazza, 1975;Azerad, 1999;Asha and Vidyavathi, 2009), requires the selection of active microorganisms (Li et al, 2008;Marvalin and Azerad, 2011).…”

“…Besides the fact that these procedures are used for the production of drug metabolites and being part of the metabolite identification process [2][3][4], this approach is increasingly showing its potentials in the extension of the so-called chemical space. Cytochrome P450 BM3 is one of the most studied bacterial P450s and has the highest catalytic activity ever detected for a monooxygenase [5].…”
