Metabolomics changes in a rat model of obstructive jaundice: mapping to metabolism of amino acids, carbohydrates and lipids as well as oxidative stress

Abstract: The study examined the global metabolic and some biochemical changes in rats with cholestasis induced by bile duct ligation (BDL). Serum samples were collected in male Wistar rats with BDL (n = 8) and sham surgery (n = 8) at day 3 after surgery for metabolomics analysis using a combination of reversed phase chromatography and hydrophilic interaction chromatography (HILIC) and quadrupole-time-of-flight mass spectrometry (Q-TOF MS). The serum levels of malondialdehyde (MDA), total antioxidative capacity (T-AOC),… Show more

“…Antioxidant defenses, as demonstrated by GSH, catalase and SOD, are found to be decreased while lipid peroxidation, as demonstrated by MDA levels, is found to be increased in rat models with OJ (7,18,19). There have been studies suggesting that LA reduced MDA levels and increased antioxidant levels such as GSH and SOD in hepatic pathologies (20)(21)(22)(23).…”

“…Compared with sham operated rats, BDL rats displayed oxidative stress, with diminished serum GSH, total antioxidant capacity, and superoxide dismutase and glutathione peroxidase activities, with upregulated serum malondialdehyde. Changes in certain amino acids, lipids, Krebs cycle intermediates, and lactic acid were signs of the effects of cholestasis on energy metabolism [98]. The BDL cholestasis rat model was shown to generate similar metabolic characteristics as thioacetamide (TAA)-induced cholestasis in rats, with excessive fatty acid oxidation, insufficient glutathione regeneration, and disturbed gut microbiota.…”

“…Val, Leu and Ile biosynthesis was observed in larvae after PCB91 exposure. Leu and Val are branched chain amino acids (BCAAs), and an appropriate ratio of BCAAs can prevent amino acid imbalance 51 . BCAAs are important amino acids in energy metabolism, and changes in BCAA levels can result in the disturbance of three major nutrients: sugar, fat and protein.…”
