Metabonomics and Toxicology

Abstract: Being an emerging field of "omics" research, metabonomics has been increasingly used in toxicological studies mostly because this technology has the ability to provide more detailed information to elucidate mechanism of toxicity. As an interdisciplinary field of science, metabonomics combines analytical chemistry, bioinformatics, statistics, and biochemistry. When applied to toxicology, metabonomics also includes aspects of patho-biochemistry, systems biology, and molecular diagnostics. During a toxicological … Show more

“…MS-based metabolomics allows targeted and untargeted metabolome analysis and it has become an indispensable tool in metabolome analysis (Milne et al, 2013 ); moreover, MS has a broader dynamic range, exhibits a high sensitivity and selectivity (Zhao and Hartung, 2015 ), and allows determining metabolite fingerprints for establishing metabolome libraries, which facilitate the identification of metabolites (Martins-de-Souza, 2014 ).…”

“…Finally, MS-based metabolomic techniques offer high selectivity and sensitivity for metabolites' identification and quantification. In fact, they are considered as the most appropriate techniques for the detection of large numbers of metabolites, and, in combination with advanced and high-throughput platforms, they may help decrease the complexity of metabolite separation (Zheng et al, 2011 ; Zhao and Hartung, 2015 ). In particular, for the total screening of the small molecules in a biological system, MS non-targeted metabolomics is a powerful tool for the identification of metabolite signals present in spectra (Naz et al, 2014 ).…”

“…In contrast, MS has a broader dynamic range and exhibits a better sensitivity and selectivity. In the past few years, the use of MS in metabolomics is rapidly growing [31] . A combination of 1 H-NMR and MS technologies would result in a better coverage of the complete metabolome.…”

“…The National Research Council Report “Toxicity Testing in the 21st Century: a Vision and a Strategy (2007)” ( NRC 2007 ) promoted the use of nonanimal models and called for embracing new technologies and basing assessments on toxicological mechanisms related to human biology and exposure. The proposed suite of tools includes the combination of computational models (integrative and targeted quantitative structure–activity predictions) and multiple in vitro assays, pathway-based approaches and toxicokinetic modeling ( Browne et al 2015 ; Bisson 2012 ; Zhao and Hartung 2015 ; Perkins et al 2014 ; Kleinstreuer et al 2014 ; Tice et al 2013 ; Kavlock et al 2012 ).…”
