Metal–organic framework-based oxygen carriers with antioxidant protection as a result of a polydopamine coating

Abstract: Rapid haemorrhage control to restore tissue oxygenation is essential in order to improve survival following traumatic injury. To this end, the current clinical standard relies on the timely administration of...

“…H 2 O 2 was further chosen as a representative free radical. Despite the relevance of such an experiment, to assess the antioxidant properties of HBOCs, very few reports have taken into consideration the biological environment. , To show the protective effect of our newly developed MPN@Hb-NPs, we first evaluated the hemolysis rate upon incubating RBCs with increasing concentrations of H 2 O 2 (Figure S2A). To demonstrate the potential of our system, the H 2 O 2 concentrations chosen were in the mM range that is much higher than the ones usually found in the body .…”

“…PDA, which is formed by the self-polymerization of dopamine in alkaline aqueous media, is an efficient adhesive material that can be deposited onto virtually any type of substrate including nanoparticulate systems. Importantly, the antioxidant properties of PDA rendered Hb-NPs that were able to scavenge the superoxide radical anion (O 2 •– ) and hydrogen peroxide (H 2 O 2 ), which are two of the most harmful ROS in our body. Additionally, these ROS scavenging properties obtained by the PDA coating translated into minimizing the autoxidation of the encapsulated Hb into metHb.…”

“…[3][4][5][6][7][8] In our group, we have recently reported the fabrication of nanoparticles (NPs) fully made of Hb by electrospray 9 or desolvation precipitation, 10,11 and also its encapsulation within poly(lactic-co-glycolic acid) (PLGA)-NPs 12,13 or metalorganic framework (MOF)-based nanocarriers (NCs). 14,15 We have demonstrated, amongst others, that the resulting HBOCs were able to reversibly bind and release oxygen over multiple rounds.…”

“…We have recently reported the encapsulation of Hb within the so-called porous coordination network (PCN)-333(Al), which is a type MOF-NCs exhibiting large void volumes, allowing for the large Hb loading that is required to achieve a high oxygencarrying capacity. 14,15 Au NZs are chosen because, to date, they are one of the most widely investigated NZs for biomedical applications. The synthesis of Au-based NZs is easy and cost-effective and allows to fine tune their morphological properties (e.g., size, shape) which, in turn, can influence the corresponding chemical and biological properties, such as catalytic activity and biocompatibility.…”
