Metastasis blood test by flow cytometry: In vivo cancer spheroids and the role of hypoxia

Abstract: Cancer hypoxia correlates with therapeutic resistance and metastasis, suggesting that hypoxic adaptation is a critical survival advantage for cancer stem cells (CSCs). Hypoxic metabolism, however, may be a disadvantage in aerobic circulation as the extremely low incidence of metastasis-compared to the high circulating tumor-cell numbers (CTCs)-appears to suggest. As rare metastatic CSCs still survive, we searched for a mechanism that protects them from oxygen in circulation. CSCs form multicellular spheroids i… Show more

“…While the migratory potential is one aspect of metastatic potential, another necessary mechanism is anchorage-independent cell survival during transport through circulation 37 . Anchorage-independent sphere formation is an indicator of the metastatic potential of a cancer cell line 38 . The sphere forming potential of the CXCR7-mutant is significantly suppressed compared to WT cells (50.5 ± 4.95 spheroids [WT] vs. 19.5 ± 2.12 spheroids [CX7-mutant]) (Fig.…”

“…VEGF is a key protein downstream of HIF-1 under hypoxic conditions. Induction of VEGF expression under hypoxic conditions causes sprouting of new blood vessels from existing endothelia, which is essential for wound repair, organ regeneration, embryonic vascular system development, and a variety of pathological conditions, including tumor angiogenesis and metastasis of various solid tumors (24,25). Treatment with sulforaphane under hypoxic conditions inhibited VEGF activity in HCT116 cells ( Fig.…”

“…These clusters also provide a special microenvironment for the tumor cells within them, and this microenvironment varies according to different sizes and constitutions of the clusters. CTC clusters with bigger sizes have significant hypoxic microenvironments compared to those with smaller sizes and single CTCs (76). …”
