Method for the Rapid Detection of SARS-CoV-2-Neutralizing Antibodies Using a Nanogel-Based Surface Plasmon Resonance Biosensor

Abstract: The efficacy of coronavirus disease 2019 (COVID-19) vaccination is closely related to the serum levels of SARS-CoV-2-neutralizing antibodies (NAb) that bind to the receptor-binding domain (RBD) of the SARS-CoV-2 spike protein. Therefore, the rapid and quantitative measurement of SARS-CoV-2 NAb in the sera of vaccinated individuals is essential to develop an effective vaccine and further achieve population immunity, that is, herd immunity. The plaque reduction neutralization test, the gold standard for NAb effe… Show more

“…In this study, we developed analytical modalities for α-thrombin detection based on its specific binding to the platelet protein GPIbα and tested different methods to investigate in-depth detection protocols via MPB and to demonstrate easy and timely detection of this candidate marker. Our nanogel platform, known for its proficiency in ligand detection such as IL-2 and SARS-COV-2 in SPR analysis, , was used by functionalizing it with GPIbα. The nSPR assay indicated that GPIbα was suitable for α-thrombin detection with not only high-fidelity binding but also wide dynamic ranges.…”

“…However, microscopic imaging lacks sensitivity for the accurate quantification of target molecules and requires sophisticated instrumentation. To overcome these limitations, bioresponsive nanogels have been employed as sensing platforms in surface plasmon resonance (SPR) analysis to investigate the MPB of programmed cell death protein 1, interleukin-2 (IL-2) receptor α, and the receptor-binding domain of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) to their ligands. …”

“…Deformable nanogels were prepared using aqueous free-radical precipitation polymerization as previously described 46, 47 . Briefly, an 88% pNIPAM/10% AAc/2% BIS solution (poly( N -isopropylacrylamide- co -acrylic acid)) was used for nanogel synthesis.…”

“…63 Functionalized NGs with the receptor-binding domain of the SARS-CoV-2 spike protein have served to detect neutralizing antibodies in the sera of individuals vaccinated against COVID-19. 66 Furthermore, hybrid NGs 67 based on Nisopropylacrylamide/N,N′-methylenebis(acrylamide) (NI-PAAM/MBA) and Cr 2 O 3 nanosheets and further decorated with AuNPs have been used to detect H 2 O 2 by surfaceenhanced Raman spectroscopy (SERS) with LOD = 10 −8 mol/ L. Also, thermoresponsive lysozyme/calcium alginate-g-poly-(NIPAAM) nanohydrogels 68 (Lys/CA-g-PNIPAAm NHs) were used to obtain a lysozyme molecularly imprinted sensor, which can selectively rebind to lysozyme in a mixture of closely related compounds. This sensor exhibited a wide linear range from 1 × 10 −9 to 1 × 10 −3 mg/mL at 30 °C.…”
