Methodology Analysis of genetic diversity in Larix gmelinii (Pinaceae) with RAPD and ISSR markers

Abstract: ABSTRACT. Dahurian larch (Larix gmelinii), a deciduous conifer, is the northernmost tree, native to eastern Siberia and nearby regions of China. We used growth traits and molecular markers to assess genetic variation in different L. gmelinii growing regions; 105 individual samples were collected from seven regions of the Qingshan Forestry Centre, Heilongjiang Province, China. The greatest genetic regional variation was seen in the Youhao area, based on coefficients of variation for tree height, diameter and vo… Show more

“…Both marker-based methods revealed the presence of abundant genetic diversity in L. gmelinii natural populations. the feasibility of the two types of markers for detecting L. gmelinii diversity was also verified, which was consistent with previous research (5,17,38).…”

“…In this study, we used random amplified polymorphic DNA (RAPD) and expressed sequence tag-simple sequence repeat (eSt-SSR) techniques to analyze the genetic diversity of 270 individual samples from six natural L. gmelinii populations (2,5,8,22,38). We compared the two types of markers with respect to testing efficiency and resolution of genetic diversity, thereby providing a theoretical basis for their effective use in the evaluation of genetic diversity and gene resources in L. gmelinii collection from the upper canopy, tender leaves were packaged, labeled, and transported on ice to the laboratory.…”

“…Dom João Hoff mann, 313, 99700-000, Erechim, RS, Brazil variability both within and among species (Rutledge et al 2000;Amin et al 2012;Cruz et al 2016). RAPD (Random Amplified Polymorphic DNA) is a molecular marker that allows detection of polymorphism without previous knowledge of genetic sequences, and is commonly used in plant population studies (Ahmed et al 2012;Zhang et al 2013;Tiwari & Shrivastava 2016). Isozymes are biochemical markers used in the investigation of genetic diversity in natural populations (Kaya & Bilben 2012;Saini & Yadav 2013), allowing analysis of genotype frequencies, coefficients of diversity genetic and heterozygosis, and estimation of possible derivations of Hardy-Weinberg´s equilibrium (Alfenas et al 1998).…”
