Methylation pattern of the putative tumor-suppressor gene LRRC3B promoter in clear cell renal cell carcinomas

Abstract: Abstract. The leucine rich repeat containing 3B (LRRC3B) gene is a putative tumor suppressor located on human chromosome 3 in the 3p24 region. LRRC3B is frequently altered in colon and gastric cancers and also in leukaemias. In this study we investigated the promoter region methylation as a possible mechanism of LRRC3B gene inactivation in clear cell renal cell carcinomas. We found that the LRRC3B gene promoter was methylated in 43% of clear cell renal carcinoma samples. However, no correlation between DNA met… Show more

“…Previous studies showed that LRRC3B is regulated by DNA methylation [44] and plays an important role in DNA repair [45]. Recent studies have proven that it could be a putative tumor suppressor gene in many types of cancer in human [46]. Interestingly, the transfection and expression of LRRC3B in gastric cancer cell line showed that immune response-related genes and IFN signaling genes can be activated effectively [47], suggesting that LRRC3B may be involved in immune-related processes that trigger the innate immune response through activation of the IFN signaling pathway.…”

“…Thus, we showed hypermethylation of the LRRC3B promoter CpG-island in 43 % of clear cell RCC's [12]. Moreover, the restoration of LRRC3B expression led to a significant decrease of colony formation by KRC/Y cell line [23].…”

“…The 7 of them were associated with other human cancers: MINT24, BHLHB2, ITGA9, RPL15, RARbeta1, RBSP3, and VHL. Some of them are accepted tumor suppressor genes, for example, LRRC3B and WNT7A [12,13].…”

“…The protein-conserved region of LRRC3B exists cAMP-and cGMP-dependent protein kinase phosphorylation sites; thus, related to cell differentiation, cycle regulation, proliferation and invasion [18,19]. Several studies have revealed LRRC3B could be as a tumor suppressor in carcinogenesis [20,21]. Formerly, the expression of LRRC3B was reduced in BC tissues and associated with tumor grade of BC [13].…”
