MHC class II cell-autonomously regulates self-renewal and differentiation of normal and malignant B cells

Abstract: Best known for presenting antigenic peptides to CD4+ T cells, major histocompatibility complex class II (MHC II) also transmits or may modify intracellular signals. Here, we show that MHC II cell-autonomously regulates the balance between self-renewal and differentiation in B-cell precursors, as well as in malignant B cells. Initiation of MHC II expression early during bone marrow B-cell development limited the occupancy of cycling compartments by promoting differentiation, thus regulating the numerical output… Show more

“…The CD11c-Cre depleted greater than 90% of Tlr9 alleles as assessed by qPCR in sorted cDCs and pDCs. As previously reported, this Cre line can be promiscuous and also targets B cells (33,36). In accordance with these data, we observed 93.2% deletion efficiency in plasmablasts, and 62.8% allele deletion in the B cell compartment.…”

“…Their EVs express immunomodulatory molecules, such as major histocompatibility complex (MHC) class I and class II molecules, whose expression levels are much higher in inflammatory conditions compared with basal conditions [68][69][70]. MHC II is essential in initiating adaptive immunity; its upregulation during B-cell development suggests its role in consolidating B-cell maturation [71]. The adaptive immune response is related to the high expression of MHC I molecules in esophageal adenocarcinoma development [72].…”

“…Transfection of Platinum-E retroviral packaging cells and retroviral transduction of target cells were performed as previously described. 71 Cell lines Murine pro-B cell leukemia cell lines B3 and F6 were originally established from transgenic mice overexpressing IL-7 under the control of the MHC II (E alpha) promoter. 35 These cells do not express endogenous MHC II or the MHC II promoter-driven IL-7 transgene.…”
