Micro‐ and Nano‐Technologies for Lipid Bilayer‐Based Ion‐Channel Functional Assays

Abstract: Ion channel proteins provide gated pores that allow ions to passively flow across cell membranes. Owing to their crucial roles in regulating transmembrane ion flow, ion channel proteins have attracted the attention of pharmaceutical investigators as drug targets for use in the studies of both therapeutics and side effects. In this review, we discuss the current technologies that are used in the formation of ion channel-integrated bilayer lipid membranes (BLMs) in microfabricated devices as a potential platform… Show more

“…6,7 More recently, mechanically stable bilayers have been be exploited, which can be formed, often semi-automatically, by virtue of advances in the bilayer formation technique. 8,9 Therefore, the design of transmembrane signaling associated with stable planar bilayer membranes will be interesting for constructing sophisticated biosensors that respond to a variety of targets.…”

“…Although the patchclamp method is considered to be the gold standard for evaluating channel activities, the method also has limitations in that the observed currents are dependent on the condition of the target cells (3). Therefore, the development of an alternate method for recording ion channel activities, complementing the patch-clamp method, would be highly desirable (5).…”

“…Reconstitution of ion channel proteins in artificial freestanding bilayer lipid membranes (BLMs) is another approach for recording ion channel activities (5,6). The BLM reconstitution system has the advantage that the researcher can control the composition of the system, including the solution phase and the lipid environment.…”

“…[8][9][10][11] Among them, electronic micro/nano sensors are particularly attractive owing to their high sensitivity response, miniaturized design, cost-effectiveness and speedy response. Field effect transistors (FETs) are surface affinity type sensors that can amplify the biological interactions into electrical signals, due to their high transconductance gain.…”
