The quest for a suitable biomaterial for medical application and tissue regeneration has resulted in the extensive research of surface functionalization of material. Poly(3-hydroxybutyrate-co-4-hydroxybutyrate) [P(3HB-co-4HB)] is a bacterial polymer well-known for its high levels of biocompatibility, non-genotoxicity, and minimal tissue response. We have designed a porous antimicrobial silver SSD blend/poly(3HB-co-4HB)-collagen peptide scaffold using a combination of simple techniques to develop a scaffold with an inter-connected microporous pore in this study. The collagen peptide was immobilised via -NH2 group via aminolysis. In order to improve the antimicrobial performance of the scaffold, silver sulfadiazine (SSD) was impregnated in the scaffolds. To confirm the immobilised collagen peptide and SSD, the scaffold was characterized using FTIR. Herein, based on the cell proliferation assay of the L929 fibroblast cells, enhanced bioactivity of the scaffold with improved wettability facilitated increased cell proliferation. The antimicrobial activity of the SSD blend/P(3HB-co-4HB)-collagen peptide in reference to the pathogenic Gram-negative, Gram-positive bacteria and yeast Candida albicans exhibited SSD blend/poly(3HB-co-4HB)-12.5 wt% collagen peptide as significant construct of biocompatible antibacterial biomaterials. Thus, SSD blend/P(3HB-co-4HB)-collagen peptide scaffold from this finding has high potential to be further developed as biomaterial.

In recent years, naturally biodegradable polyhydroxyalkanoate (PHA) monopolymers have become focus of public attentions due to their good biocompatibility. However, due to its poor mechanical properties, high production costs, and limited functionality, its applications in materials, energy, and biomedical applications are greatly limited. In recent years, researchers have found that PHA copolymers have better thermal properties, mechanical processability, and physicochemical properties relative to their homopolymers. This review summarizes the synthesis of PHA copolymers by the latest biosynthetic and chemical modification methods. The modified PHA copolymer could greatly reduce the production cost with elevated mechanical or physicochemical properties, which can further meet the practical needs of various fields. This review further summarizes the broad applications of modified PHA copolymers in biomedical applications, which might shred lights on their commercial applications.

The rate and extent of microbial polyhydroxyalkanoates (PHAs) production rely on the availability of substrates, growth of microbial biomass, and intracellular accumulation of polymer under nitrogen‐limited conditions. The dynamics of PHAs production captured through various structured or unstructured models can be extended to design an optimal feeding strategy for process intensification. Large variability in process assumptions, choices of kinetics, and model complexity is expected depending on substrate(s), microbial metabolism, and discretization of the process under consideration. This communication attempts to review the estimation of stoichiometric yield coefficients, metabolic modelling, and choices of unstructured kinetics in microbial PHA production. Implementational irregularities in parameter estimation and quality check in modelling exercises have also been reviewed. It is observed that the scope of the majority of the “modelling” studies is confined to the estimation of stoichiometric parameters with limited utility. In dynamic models, microbial growth is often described using either Monod or logistic variants, while PHAs production adopts a Luedeking–Piret expression with or without substrate inhibition. Though model selection, regression with experimental data, parameter estimation, and model validation are integral parts of the exercise, very few provide sufficient coverage on all those aspects. Application of the model to control or optimize the bioprocess has rarely been attempted.
