Filtros lentos de areia (FL) são amplamente aplicados para tratar a água para uso potável. A remoção de contaminantes (matéria orgânica e microrganismos) ocorre principalmente na camada de biofilme, enriquecida de comunidade microbiana (CB), são ainda pouco compreendidos. O presente trabalho é uma revisão sistemática, com objetivo de analisar e identificar artigos sobre CB em FL, com base nas plataformas Scopus, Web of Science e ScienceDirect e retorno de 2496 artigos. Destes, apenas 22 satisfizeram os critérios de inclusão, analisados integralmente. A avaliação da qualidade metodológica dos estudos incluídos identificou 10 filos, 20 gêneros e 6 técnicas de identificação (T). Assim, o estudo demonstrou a variedade de microrganismos e respectivas T, além de mostrar a necessidade de pesquisas com maior rigor de T e com evidências sobre a função de cada microrganismo.

Affected by the complex operation mode of strong injection and strong production, sand production can seriously affect the life cycle and peak shaving capacity of gas storage. In this paper, combined with the actual production situation of China’s largest gas storage (H Gas Storage), the effect of the gas flow rate, production pressure difference, formation pressure drop, permeability, and water saturation on sand production was systematically analyzed via an indoor sand production simulation experiment. The results showed that from the initial flow rate of 1.88 L/min to a critical flow rate of 3.87 L/min, the core permeability and the stage sand production continued to increase; however, during the flow rate from 3.87 to 8.58 L/min, the core permeability and the stage sand production decreased gradually. The whole process showed that only a small amount of free sand was produced in the early anhydrous production stage. Proper sand production in this stage can make the rock permeability and the gas production grow to a certain extent. But when the gas flow rate reached 3.87–8.58 L/min, pore throats were blocked by sand particles of large sizes. When all sand particles were carried away by the gas flow, no sand production occurred and the rock permeability remained unchanged. In this experiment, the critical pressure difference was 5 MPa. Under the same production pressure difference, the greater the rock permeability, the greater the stage sand production; similarly, under the same rock permeability, the greater the production pressure difference, the greater the stage sand production. In the case of high rock permeability, sand production occurred when the displacement pressure difference was 2 MPa. For the formation pressure drop, sand production occurred when the effective stress reached 10 MPa; furthermore, when the effective stress reached 16.5 MPa, the stage sand production reached the largest value; finally, when the effective stress reached 28 MPa, no sand production occurred anymore. Sand production was easy to occur in cores containing water, which was related to the hydration and swelling of clay minerals, the increase of seepage resistance in gas–water two-phase fluids, and the increase of shear stress in pore throats.

Nitrogen-metabolising Microorganism Analysis in Rapid Sand Filters From Drinking Water Treatment Plant by Culturing and Metagenomics

Sand filter (SFs) are common treatment processes for nitrogen pollutants removal in drinking water treatment plants (DWTPs). However, the mechanisms on the nitrogen-cycling role of SFs are still unclear. In this study, 16S rRNA gene amplicon sequencing was used to characterise the diversity and composition of the bacterial community in SFs from DWTPs. Additionally, metagenomics approach was used to determine the functional microorganisms involved in nitrogen cycle in SFs. Our results showed that Proteobacteria, Acidobacteria, Nitrospirae, and Chloroflexi dominated in SFs. Subsequently, 85 high-quality metagenome-assembled genomes (MAGs) were retrieved from metagenome datasets of selected SFs involving nitrification, assimilatory nitrogen reduction, and denitrification processes. Read mapping to reference genomes of Nitrospira and the phylogenetic tree of the ammonia monooxygenase subunit A gene, amoA, suggested that Nitrospira is abundantly found in SFs. Furthermore, according to their genetic content, a nitrogen metabolic model in SFs was proposed using representative MAGs and pure culture isolates. Quantitative real-time polymerase chain reaction (PCR) showed that ammonia-oxidising bacteria (AOB) and archaea (AOA), and complete ammonia oxidisers (comammox) were ubiquitous in the SFs, with the abundance of comammox being higher than that of AOA and AOB. Moreover, we identified a bacterial strain with a high NO3-N removal rate as Pseudomonas sp., which could be applied in the bioremediation of micro-polluted drinking water sources. Our study provides insights into functional nitrogen-metabolising microbes in SFs of DWTPs.
