Variation of soil microbial community abundance and structure has great implications for soil fertility and nutrient cycling. A better understanding of soil microbial community dynamics under different land use types is undoubtedly needed in order to develop sustainable land use schemes. The current study aimed to assess how soil microbial community changed after replacement of maize–soybean crop by sugarcane, mulberry, or forage grass crop in a karst area of Southwest China. Mature forests were included for comparison. Phospholipid fatty acid (PLFA) method was used to characterize soil microbial community abundance and structure. The abundances of total PLFAs and PLFAs of bacteria, fungi, actinomycetes, and arbuscular mycorrhizal fungi were significantly increased in the forage grass field but not in the sugarcane and mulberry fields relative to the maize–soybean field. Total PLFAs' abundance in the forage grass field was increased by 81% compared with that in the maize–soybean field but was about 52% lower than that in the forest. The microbial community structure was not distinguished as much as the microbial abundance among the five land use types. Soil organic carbon (SOC) was identified as the primary factor affecting both soil microbial abundance and structure. Soil microbial community abundance was positively correlated with SOC, but the ratios of fungal to bacterial PLFAs and Gram‐positive to Gram‐negative bacterial PLFAs were negatively correlated with SOC. Our findings suggest that the replacement of the maize–soybean rotation system by forage grass cultivation has the potential to improve soil fertility in the karst region, Southwest China.

Summary Profiling of microbial communities in environmental samples often utilizes phospholipid fatty acid (PLFA) analysis. This method has been used for more than 35 years and is still popular as a means to characterize microbial communities in a diverse range of environmental matrices. This review examines the various recent applications of PLFA analysis in environmental studies with specific reference to the interpretation of the PLFA results. It is evident that interpretations of PLFA results do not always correlate between different investigations. These discrepancies in interpretation and their subsequent applications to environmental studies are discussed. However, in spite of limitations to the manner in which PLFA data are applied, the approach remains one with great potential for improving our understanding of the relationship between microbial populations and the environment. This review highlights the caveats and provides suggestions towards the practicable application of PLFA data interpretation.

MICROBIAL METABOLOMICS: INNOVATIONS AND APPLICATIONS. Over the past few years, metabolomics has been employed in a broad range of applications in different research fields. Microbial metabolomics has been applied in several microbiological areas, such as identification of microorganisms, cell mutations, functional gene approach, identification of unique metabolic pathways and microbial engineering, leading to a better understanding of the global metabolism and metabolic regulation of certain systems. Metabolites produced by microorganisms constitute a large promising group with diverse applications; however, most of these systems are still unknown and underexplored. In this context, the overall analysis of metabolites involved in a biological system can be used to discover potential biomarkers or specific metabolic changes due to a biological phenomenon studied. In this sense, this review aims to understand the main challenges faced by each step of the workflow in the metabolic analysis of microorganisms and how these difficulties have been overcome. A critical analysis of the articles published within the last five years will be accomplished in this paper to understand how these studies have contributed to the microbial metabolomics research. Moreover, major trends observed in this area will be highlighted here in order to guide readers interested in microbial metabolomics in their future researches. Tabela 2.
