Microneedle-array patches loaded with hypoxia-sensitive vesicles provide fast glucose-responsive insulin delivery

Abstract: A glucose-responsive "closed-loop" insulin delivery system mimicking the function of pancreatic cells has tremendous potential to improve quality of life and health in diabetics. Here, we report a novel glucose-responsive insulin delivery device using a painless microneedle-array patch ("smart insulin patch") containing glucoseresponsive vesicles (GRVs; with an average diameter of 118 nm), which are loaded with insulin and glucose oxidase (GO x ) enzyme. The GRVs are self-assembled from hypoxia-sensitive hyalu… Show more

“…[10] Alternatively, in consideration of the essential role of glucose in providing energy for tumor metabolism, [11] we strategically starved the tumors by depleting the intratumoral glucose by a glucose-metabolic reaction, which oxidized glucose into gluconic acid and H 2 O 2 using the catalysis of glucose oxidase (GOx). [12] As we know, H 2 O 2 causes malignant transformation of normal cells at endogenous concentrations, [13] but in turn triggers cancerous cell death at high concentrations.…”

“…Moreover, microneedle devices can be designed to dissolve in the skin, eliminating the issue of microneedle remaining and removal from the skin and allowing a safe disposal without biohazardous waste. 35 Although microneedles show great promise for the delivery of drugs 36 and also as sensing devices, 37 it is in the research area of vaccines that they showed, so far, more advances. It was demonstrated that vaccination using microneedles triggers stronger immune responses comparing to conventional injection procedures, allowing sparing of antigens.…”

“…One option involves a micro-needle patch that penetrates the outer surface of the skin. [47,48] With this device, insulin is delivered through prongs on the patch and is released in proportion to glucose levels. This single disposable device consists of a 6 mm 2 patch with 121 microneedles embedded in it.…”
