MicroRNAs regulate tight junction proteins and modulate epithelial/endothelial barrier functions

Abstract: Tightly controlled epithelial and endothelial barriers are a prerequisite for life as these barriers separate multicellular organisms from their environment and serve as first lines of defense. Barriers between neighboring epithelial cells are formed by multiple intercellular junctions including the ‘apical junctional complex—AJC’ with tight junctions (TJ), adherens junctions (AJ), and desmosomes. TJ consist of tetraspan transmembrane proteins like occludin, various claudins that directly control paracellular … Show more

“…Finally, microRNAs (miRNAs) post-transcriptionally regulate TJ formation and barrier function 67. Indeed, different miRNAs have been shown to modulate CLDN1 expression (table 2): targeting of CLDN1 mRNA by miR-29 has been shown to regulate intestinal permeability,68 while the regulation of CLDN1 mRNA by miR-155 plays an important role in promoting colorectal cancer (CRC) cell migration and invasion 69.…”

“…In this setting, miR-155, miR-320, miR-126, and miR-222 have been shown to regulate adherens junction disassembly, cell migration, and cell morphology, which contribute to changes in vascular permeability [102,103]. Upon VEGF stimulation, the expression of miR-125b has been shown to be transiently induced in endothelial cells which in turn down-regulates the translation of vascular endothelialcadherin messenger RNA (mRNA) [67] and thus affecting normal barrier function [67,101].…”

“…Tissue barriers include tight junctions (TJs), adherens junctions, and desmosomes, which regulate the paracellular permeability of epithelial layers across the apical/ basolateral axis [20] . The main protein complexes of TJs are composed of transmembrane proteins such as claudins, occludin and junctional adhesion molecules [21] .…”
