Summary 1.Urban and rural populations of animals can differ in their behaviour, both in order to meet their ecological requirements and due to the constraints imposed by different environments. The study of urban populations can therefore offer useful insights into the behavioural flexibility of a species as a whole, as well as indicating how the species in question adapts to a specifically urban environment. 2. The genetic structure of a population can provide information about social structure and movement patterns that is difficult to obtain by other means. Using non-invasively collected hair samples, we estimated the population size of Eurasian badgers Meles meles in the city of Brighton, England, and calculated population-specific parameters of genetic variability and sex-specific rates of outbreeding and dispersal. 3. Population density was high in the context of badger densities reported throughout their range. This was due to a high density of social groups rather than large numbers of individuals per group. 4. The allelic richness of the population was low compared with other British populations. However, the rate of extra-group paternity and the relatively frequent (mainly temporary) intergroup movements suggest that, on a local scale, the population was outbred. Although members of both sexes visited other groups, there was a trend for more females to make intergroup movements. 5. The results reveal that urban badgers can achieve high densities and suggest that while some population parameters are similar between urban and rural populations, the frequency of intergroup movements is higher among urban badgers. In a wider context, these results demonstrate the ability of non-invasive genetic sampling to provide information about the population density, social structure and behaviour of urban wildlife.

Genetic structure of the European polecat (Mustela putorius) and its implication for conservation strategies

During the last century, the European polecat Mustela putorius populations in most of Europe declined and survived in fragmented patches, because of habitat alterations and direct persecution. To assess the genetic consequences of the demographic decline and to describe the spatial pattern of genetic diversity, 250 polecats sampled at seven localities from five European countries -Poland, Denmark (southern Denmark and northern Denmark), Spain, Belgium (eastern and western) and the Netherlands -were screened by means of nine microsatellite loci. Genetic diversity estimated by mean expected heterozygosity (H E ) and allelic richness (AR) were moderately high within populations [range: 0.50 (northern Denmark) H E 0.64 (Poland) and 1.33 AR 7.80] as compared with other carnivores and mustelids. Bottleneck tests suggested that polecat populations in southern Denmark and Poland have declined recently and populations from northern Denmark and the Netherlands have expanded recently, whereas the remaining populations did not show any sign of demographic change. Recent demographic changes could suggest that some of the populations are still not in equilibrium, which could partly explain the relatively high genetic variability observed in polecat populations despite the drastic decline in population size observed in several European countries. A significant heterozygote deficiency [F IS = 0.19; 0.01 95% confidence interval (CI) 0.32] suggests substructuring within the total European sample. Partitioning of the genetic variation among sampling locations (F ST = 0.14; 0.06 95% CI 0.23) and pairwise F ST between localities (range: 0.01 F ST 0.37) without any correlation with the geographic distances between localities were found, suggesting a recent divergence and a restriction of gene flow between populations and the action of genetic drift. An assignment test showed that the Polish and the northern Danish populations were the most unique, whereas the other populations were partially admixed. Factorial component analysis tests indicate a subdivision of the total sample into two distinct groups: one including the samples from Poland and the two Danish localities and the second group comprising the remaining localities investigated. The observed pattern of genetic differentiation is suggested to be due to two main routes of recolonization after the last glacial period. To compare the results obtained with microsatellite data, the most variable region of the mitochondrial DNA (d-loop) was sequenced and different phylogenetic reconstructions and genetic diversity analyses based on nucleotide (p) and haplotype diversity (h) measures within populations were performed using a subsample of populations. Especially in urbanized landscapes, habitat fragmentation and increasing numbers of infrastructural features may limit genetic exchange among wildlife populations. Yet, whether this results in genetic differentiation among individuals in different habitat fragments will depend on both the species studied and the composition of the landscape. European pine martens (Martes martes) show clear spatial structure at a Europe-wide scale, but whether gene flow among habitat patches can be maintained at a more local scale in intensively urbanized areas remained unclear. Here, we analysed genotypic data from 270 pine martens sampled from locations scattered across the Netherlands, one of the most densely populated areas in the world. Using Bayesian clustering models we show that most likely maximum two large subpopulations exist in the Netherlands. We observed relatively low levels of genetic differentiation and genetic evidence of regular long-distance dispersal by juveniles that must have crossed one or multiple major highways. Our results suggest that genetic exchange among Dutch pine martens has, until 2010, not been impacted severely by the countries' dense infrastructural network. Furthermore this species seems to have maintained its genetic diversity despite a recent demographic bottleneck. These conclusions support the idea that the effects of habitat fragmentation may strongly differ between (groups of) species, and that prioritization and optimization of management decisions thus requires direct study of the targeted species.
