Microsatellite Status and IκBα Expression Levels Predict Sensitivity to Pharmaceutical Curcumin in Colorectal Cancer Cells

Abstract: Clinical utilization of curcumin in colorectal cancer (CRC) was revived as a result of the development of novel curcumin formulations with improved bioavailability. Additionally, identification of biomarkers for curcumin sensitivity would also promote successful clinical applications. Here, we wanted to identify such biomarkers in order to establish a predictive model for curcumin sensitivity. Thirty-two low-passage CRC cell lines with specified tumor characteristics were included. Curcumin suppressed cell pro… Show more

“…The highest incidence and mortality rates of 52.3% and 54.2% were observed in Asia (9). The 5year prevalence has been recorded in 5,253,335 persons across all populations (10). Furthermore, CRC is more common in men (23.4) than women (16.2) according to agestandardized incidence rates calculated per 100,000 persons (11).…”

“…Another difference is the microsatellite stability; in fact, as reported by Yashiro et al, LoVo has an MSI phenotype, while HT-29 cells do not [84]. Some of these features may influence the response to SFN treatments, as in the case of other phytochemical compounds such as curcumin, the activity of which may be affected by the microsatellite status [85].…”

“…Investigators screened different molecular types of CRC on cell lines to find the ones with the highest sensitivity to curcumin, a natural product [ 34 ]. Most curcumin-sensitive cell lines were of the MSS molecular type, and they had high baseline levels of the IκBα protein.…”

“…Most curcumin-sensitive cell lines were of the MSS molecular type, and they had high baseline levels of the IκBα protein. Conversely, curcumin-resistant cell lines were mainly of the MSI type, and showed low baseline IκBα levels [ 34 ]. This suggests curcumin could be useful for treatment or prevention of MSS CRC, and there are ongoing efforts to increase its bioavailability [ 35 ].…”
