Microwave‐assisted extraction in combination with HPLC‐UV for quantitative analysis of six bioactive oxoisoaporphine alkaloids in Menispermum dauricum DC

Abstract: A novel and reliable method based on microwave-assisted extraction (MAE) followed by HPLC-UV was developed and validated for the simultaneous quantification of six pharmacologically important oxoisoaporphine alkaloids in the total plants of Menispermum dauricum DC. The optimal MAE extraction condition was performed at 60°C for 11 min with ethanol-water (70:30, v/v) as the extracting solvent, and the solvent to solid ratio was 20:1. Chromatographic separation was achieved on a reversed-phase YMC C18 column (250… Show more

“…It is recommended that these homogenization procedures be performed in liquid nitrogen in order to avoid degradation. After homogenization, several selective metabolite extraction methods, including microwave-assisted extraction (Teo et al 2013 ; Gemperline et al 2016b ; Wei et al 2016 ) , ultrasound-assisted extraction (Chemat et al 2017 ), high voltage electric discharge extraction (Li et al 2019 ), supercritical fluid extraction (SFE) (Gallego et al 2019 ), enzyme-assisted extraction (Puri et al 2012 ), and solid-phase extraction (SPE) (Reyes-Garcés and Gionfriddo 2019 ) are commonly used in combination or sequentially. Notably, given the fact that SFE affords various advantages such as chemically stable, environmentally friendly, low toxicity, and not flammable, it has been chosen as a good extraction strategy for volatile compounds (e.g., terpenes and aromatic compounds) from plant samples (Naz et al 2017 ).…”

“…The standard of DMP was isolated in the author's laboratory. The procedure for isolation, purification, and characterization of DMP as well as the corresponding spectral and physicochemical data (MS, 1 H-NMR, and 13 C-NMR) have been published by the author (Shao et al, 2019;Wei et al, 2016). Based on these data as well as the literature report (Li et al, 2013), the structure of DMP was accurately characterized.…”

“…Isoquinoline alkaloid oxoisoaporphines (OAs, Fig. 1) isolated from Menispermum dauricum DC roots (family Menispermaceae) [21][22][23]. Menispermum dauricum DC roots as a perennial herb distributed in the northeast, north and east of China, southern Japan, Korea and Russian Siberia widely has been used as traditional medicine treatment for analgesic and antipyretic [24].…”
