Migration of human intestinal lamina propria lymphocytes, macrophages and eosinophils following the loss of surface epithelial cells

Abstract: Lymphocytes and macrophages are present in the normal intestinal lamina propria, separated from the epithelial monolayer by the basement membrane. There is evidence for movement of mononuclear cells through the lamina propria, entering from the systemic circulation and exiting via lymphatic channels. The goal of our studies was to investigate the capacity of cells to migrate out from the lamina propria into the lumen following the loss of surface epithelial cells. An in vitro model was therefore established in… Show more

“…Intestinal samples were then denuded of epithelium and cultured to allow migration of cells from the LP. Consistent with a previous study [9], 75% of the cells which migrated from the LP, after 36h, were CD3+ T cells and of these 55% expressed CD4. However, we show for the first time that the CD4+ population within the LP also expressed IL-36R.…”

“…The mucosal samples were washed thoroughly with calcium-and magnesium-free Hanks balances salt solution (HBSS; Gibco BRL, Gaithesburg, MD, USA), to remove adherent luminal material. Lamina propria cells were isolated from mucosal samples using a previously described technique [9]. table 1and as previously reported [12][13] and PCR product obtained was also shown to be of correct size as that previously reported [12].…”

“…Primary intestinal myofibroblast cultures were established from histologically normal margins of surgically resected colonic or ileal tissue by using the outgrowth method described by Mahida et al (29). All other cells were obtained from American Type Culture Collection (Manassas, VA).…”

“…18Co was used in the study as a representative CMF. Additional primary cultures of subepithelial CMFs were generated according to the method described by Mahida et al (37,38). The purity of isolated cells was analyzed by immunohistochemistry and flow cytometry analysis as described below.…”
