miR-122 affects the viability and apoptosis of hepatocellular carcinoma cells

Abstract: OBJECTIVE. miR-122 is highly abundant in liver and a hepato-specific microRNA. There is evidence to show that miR-122 expression is down-regulated in human hepatocellular carcinoma (HCC). It is not known whether miR-122 affects the cellular behavior of hepatoma cells. The aim of this study was to investigate the effects of miR-122 on the viability and apoptosis of hepatoma cells. MATERIAL AND METHODS. The viability and apoptosis of Huh-7 and HepG2 cells treated with miR-122 or miR-122 antisense RNA (anti-miR-1… Show more

“…Similar findings have been obtained by other research groups (15,16). In addition, miR-122 has been shown to influence apoptosis; transfections of the hepatoma cell line Huh-7 with a miR-122 mimic produced an up-regulation in apoptosis levels as indicated by both flow cytometry and TUNEL assay (17).…”

“…No significant alterations were found in the proliferation of Huh7, HepG2, and QSG-7701 cells treated with 5-aza-dc in the present study; however, miRNA-122 expression levels were significantly elevated, which is less consistent with previous reports (Gramantieri et al, 2007;Wu et al, 2009). The reasons for this difference may be associated with the low levels of miRNA-122 expression in the 3 hepatic cell lines themselves; the levels of miRNA-122 elevation were limited after demethylation treatment and insufficient to induce cell proliferation-related changes.…”

“…miR-122 is a hepatic tumor suppressor miR-122 levels are reduced in experimental models and human samples of HCC, and loss of miR-122 is associated with tumor invasiveness and cancer progression (13)(14)(15). Therefore, it has been speculated that this miR acts as a tumor suppressor.…”
